In Arabidopsis root meristem, regulation of protein movement has been shown to be important for positional information and stem cell maintenance. These processes are mainly controlled by protein interactions and transcriptional activation. The cell fate determinant SHORTROOT (SHR) moves between cell layers in the root and activates its target and binding partner SCARECROW (SCR). Together they promote asymmetric cell division in the root meristem. SHR movement is regulated by JACKDAW (JKD) and BALDIBIS (BIB), two C2H2 zinc finger proteins (named here BIRD proteins).

Our data indicate that both JKD and BIB are able to restrict SHR movement by nuclear retention through protein-protein interactions. To understand the spatiotemporal protein dynamics at different developmental stages, we optimized FLIM—FRET techniques in living Arabidopsis roots. Here we show that SCR and SHR interact in specific cell types. Analysis of interactions between BIRD proteins and SCR/SHR will bring more insights into how this complex network regulates SHR movement and asymmetric cell division in the Arabidopsis root meristem.
